چکیده

Prostaglandin E2 (PGE2) is an important modulator of cytokine-driven inflammation. Using GeneChip analysis, we found that interleukin (IL)-1β induces the gene expression of PTGER4, which encodes the PGE2 receptor subtype EP4 (PGE2EP4). This subtype is one of four PGE2 receptors occurring in synoviocyte MH7A cells. Immunofluorescence microscopy revealed a corresponding upregulation in the production of PGE2EP4 protein in IL-1β-pretreated MH7A cells. PGE2 alone has no effect on IL-8 production, but in cells pretreated with IL-1β it markedly enhances IL-8 production. Moreover, a stimulatory effect of PGE2 on IL-8 production in the synoviocyte MH7A cells was observed. These results indicate that, in the synovial tissues of patients with rheumatoid arthritis, PGE2 stimulates the release of IL-8 from the fibroblastic cells classified as present, thereby exacerbating inflammation.

Prostaglandin E2 regulates Th17 cell differentiation and function through cyclic AMP and EP2/EP4 receptor signaling

Prostaglandins, particularly prostaglandin E2 (PGE2), play an important role during inflammation. This is exemplified by the clinical use of cyclooxygenase 2 inhibitors, which interfere with PGE2 synthesis, as effective antiinflammatory drugs. Here, we show that PGE2 directly promotes differentiation and proinflammatory functions of human and murine IL-17-producing T helper (Th17) cells. In hum...

Genistein suppresses tumor necrosis factor α-induced inflammation via modulating reactive oxygen species/Akt/nuclear factor κB and adenosine monophosphate-activated protein kinase signal pathways in human synoviocyte MH7A cells

AIMS Genistein, an isoflavone derivative found in soy, is known as a promising treatment for rheumatoid arthritis (RA). However, the detailed molecular mechanism of genistein in suppression of proinflammatory cytokine production remains ambiguous. The aim of this work was to evaluate the signal pathway by which genistein modulates inflammatory cytokine expression. MATERIALS AND METHODS MH7A c...

Prostaglandin E2 produced by Entamoeba histolytica binds to EP4 receptors and stimulates interleukin-8 production in human colonic cells.

Entamoeba histolytica pathogenesis in the colon occurs in a stepwise fashion. It begins with colonization of the mucin layer, which is followed by stimulation of a proinflammatory response that causes nonspecific tissue damage that may facilitate parasite invasion of the underlying colonic mucosa. Unfortunately, the parasite and/or host factors that stimulate a proinflammatory response in the g...

Doxycycline Attenuates Endotoxin-Induced Uveitis by Prostaglandin E2-EP4 Signaling.

PURPOSE We explored the anti-inflammatory effects of doxycycline in experimental uveitis and the underlying mechanisms. METHODS Rats with endotoxin-induced uveitis (EIU) received doxycycline (1.5 mg/kg) or the control vehicle via intraperitoneal injection. Clinical scores were graded under a slit lamp. Rat peritoneal macrophages were used in vitro to further explore the anti-inflammatory mech...

Interleukin-1beta-induced mucin production in human airway epithelium is mediated by cyclooxygenase-2, prostaglandin E2 receptors, and cyclic AMP-protein kinase A signaling.

We reported recently that interleukin (IL)-1beta exposure resulted in a prolonged increase in MUC5AC mucin production in normal, well differentiated, human tracheobronchial epithelial (NHTBE) cell cultures, without significantly increasing MUC5AC mRNA (Am J Physiol 286:L320-L330, 2004). The goal of the present study was to elucidate the signaling pathways involved in IL-1beta-induced MUC5AC pro...
